Automated analysis and sorting of human induced pluripotent stem cell (hiPS) clusters using large particle flow cytometry

Physical properties like size, optical density and fluorescence can be traced back for every dispensed cluster. When combined with a fluorescence microscope, correlation between the profile and the micrograph can be made for each individual cluster. An example of this type of analysis showed that cluster clonality is maintained. Clusters carrying either a green or red fluorescent tag were mixed and incubated, allowing time for rearrangement or migration of the cells between clusters. However, analysis of these clusters indicated that clonality was preserved during the sorting process and clusters remained either all green or all red.
